ABSTRACT

ObjectiveTo investigate the mechanism of Zuogui Jiangtang Qinggan prescription (ZJQP) in improving glucose and lipid metabolism in loss of skeletalmuscle-specific insulin-like growth factor-1 receptor function (MKR) mice with type 2 diabetes mellitus (T2DM) and non-alcoholic fatty liver disease (NAFLD). MethodNAFLD was induced by high-fat diet feeding for 8 weeks in MKR mice, which were randomly divided into model group, metformin group (0.067 g·kg-1), and ZJQP high and low-dose groups(14.8, 7.4 g·kg-1). Ten FVB mice of the same age were used as the normal group. After 8 weeks of drug treatment, the oral glucose tolerance test (OGTT) was performed, the serum was taken to detect triacylglycerol (TG) and total cholesterol (TC), and the wet weight of the mouse liver was weighed. Haematoxylin-eosin (HE) staining and oil red O staining were performed to assess histopathology of liver. The mRNA expression and protein expression of Fork head box protein O1 (FoxO1), phosphoenolpyruvate carboxykinase (PEPCK), glucose-6-phosphatase (G6Pase), and apolipoprotein C3 (ApoC-Ⅲ) in liver tissues were detected by real-time fluorescent quantitative PCR (Real-time PCR) and Western blot, respectively. ResultAs compared with the normal group, the levels of fasting blood glucose, liver index, serum TG, TC, and OGTT of mice in the model group increased significantly (P<0.01). As compared with model group, the fasting blood glucose and liver index of the mice in the metformin group and the ZJQP group decreased significantly (P<0.01), the serum levels of TG and TC in the high-dose ZJQP group decreased significantly (P<0.05,P<0.01), and the OGTT of mice in the metformin group and the high-dose ZJQP group improved (P<0.05). In histopathology, as compared with the normal group, mice in the model group showed decreased lipid droplets and vacuoles in hepatocytes, and their volumes became larger. Compared with the model group, the ZJQP group and metformin group showed that the lipid droplets in liver tissues were reduced, the vacuoles in liver cells were reduced, and the volume was smaller. At the molecular level, as compared with the normal group, the mRNA and protein levels of FoxO1, PEPCK, G6Pase, and ApoC-Ⅲ in liver tissues of mice in the model group were significantly up-regulated (P<0.01). As compared with the model group, the mRNA and protein levels of FoxO1, PEPCK, G6Pase, and ApoC-Ⅲ in the ZJQP group was significantly decreased (P<0.01). ConclusionZJQP can improve the glucose and lipid metabolism of T2DM with NAFLD and repair the pathological damage of liver, which may be through regulating the expression of FoxO1, PEPCK, G6Pase, ApoC-Ⅲ-related proteins in liver tissues to achieve the effects of regulating lipid, lowering glucose, and delaying hepatic steatosis.

ABSTRACT

Objective:To investigate the action mechanism of Yinchenhao Tang against type 2 diabetes mellitus (T2DM) complicated with non-alcoholic fatty liver disease (NAFLD) in MKR mice. Method:Forty eight-week-old MKR mice were fed a high-fat diet for eight weeks and then divided into the model group,original Yinchenhao Tang (17.16 g·kg<sup>-1</sup>) group,Yinchenhao Tang group at a specified dose (4.68 g·kg<sup>-1</sup>) in teaching materials,and positive drug [metformin + simvastatin, (65+2.6)×10<sup>-3</sup> g·kg<sup>-1</sup>] group. Another 10 MKR mice of the same age were classified into the blank group and 10 FVB mice into the normal group. After eight weeks of intragastric administration in each group,the liver wet weight,oral glucose tolerance test (OGTT),serum inflammatory factors interleukin-6 (IL-6) and tumor necrosis factor-<italic>α</italic> (TNF-<italic>α</italic>),and changes in blood lipid and liver function were determined. Hematoxylin-eosin (HE) staining was conducted for observing the morphological changes in liver tissue under a transmission electron microscope,followed by the detection of Toll-like receptor 4 (TLR4),myeloid differentiation factor 88 (MyD88),and nuclear transcription factor-<italic>κ</italic>B (NF-<italic>κ</italic>B) protein expression by Western blot. Result:Compared with the model group,the medication groups exhibited significantly reduced liver wet weight index (<italic>P</italic><0.01),improved OGTT result (<italic>P</italic><0.05),and down-regulated serum IL-6 and TNF-<italic>α</italic> levels (<italic>P</italic><0.01). In terms of morphological changes,Yinchenhao Tang protected the hepatocyte structure and alleviated hepatocyte steatosis. Moreover, Yinchenhao Tang obviously down-regulated the protein expression levels of TLR4,MyD88,and NF-<italic>κ</italic>B in liver tissue of MKR mice with T2DM combined with NAFLD (<italic>P</italic><0.05),and the down-regulation of TLR4 and NF-<italic>κ</italic>B in the original Yinchenhao Tang group was better than that in the Yinchenhao Tang group at a specified dose in teaching materials (<italic>P</italic><0.05). Conclusion:Yinchenhao Tang is able to reduce inflammatory factor levels and down-regulate TLR4,MyD88,and NF-<italic>κ</italic>B expression in liver tissue to relieve the pathological liver injury and interfere with T2DM combined with NAFLD of MKR mice. It exerts a certain liver-protective effect by lowering the blood lipids and delaying the hepatic inflammation.

ABSTRACT

Objective: To observe the effect of Baihu and Renshen Decoction on intestinal barrier and TLR4/NF-κB inflammatory signaling pathway in type 2 diabetes MKR mice. Methods: A total of 32 MKR mice were randomly divided into model group, low dose Baihu and Renshen Decoction group, high dose Baihu and Renshen Decoction group and metformin group after 4 weeks with the high-fat diet. Another eight FVB/N mice were served as control group. After 4-week intragastric administration, fasting blood glucose (FBG), oral glucose tolerance test (OGTT), the levels of interleukin-6 (IL-6), interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α) and lipopolysaccharide (LPS) in serum were examined. The morphological changes of colon tissue were observed by hematoxylin-eosin (HE) staining. The mRNA expression of TLR4, NF-κB, ZO-1, Claudin-1 and Occludin were measured by qRT-PCR method. The protein expression of ZO-1, Claudin-1 and Occludin in colon were measured by Western blotting. Results: Compared with model group, FBG in each administration group were significantly decreased (P < 0.01), serum IL-6, IL-1β, TNF-α, LPS and TLR4, NF-κB mRNA expressions were decreased (P < 0.05, P < 0.01) in high dose Baihu and Renshen Decoction group and metformin group. The mucosal epithelium integrity was improved. The mRNA and protein expression levels of ZO-1, Claudin-1, Occludin were increased (P < 0.05, P < 0.01) in high dose Baihu and Renshen Decoction group and metformin group; The mRNA expression levels of ZO-1, Claudin-1, Occludin were increased (P < 0.05) in low dose Baihu and Renshen Decoction group, while protein expressions showed no statistical difference. Conclusion: Baihu and Renshen Decoction can consequently reduce the levels of LPS and pro-inflammatory cytokines in MKR mice, regulate TLR4/NF-κB signaling pathway. It can significantly improve insulin resistance and reduce blood glucose levels by improving endotoxaemia, ameliorating intestinal injury and attenuating inflammation.

ABSTRACT

Objective To investigate the specific expression of endoplasmic reticulum stress-related GRP78 and CHOP in podocyte of MKR mice with diabetic nephropathy (DN);To discuss the intervention effect of Zuogui Jiangtang Yishen Decoction (ZGJTYS). Methods 8-week old MKR mice were randomly divided into five groups:blank group, model group, ZGJTYS group, 4-Phenylbutyric acid group, gliquidone+benazepril group, and normal control group consisting of wild type C57BC/6 mice, 10 mice per group. All mice from model group and each treatment group received high-fat diet feed and unilateral nephrectomy to make the DN model. Mice from treatment groups received medicine intervention for four weeks. The levels of FBG were detected by electrochemical detection method, and the UmAlb was detected by ELISA. The expressions of GRP78, CHOP mRNA, and protein were detected by RT-PCR and Western blot. The morphological structure changes of the podocytes were observed by transmission electron microscopes. Results Compared with blank group, FBG and UmAlb in the model group increased significantly (P<0.01);the expressions of GRP78 mRNA and protein markedly decreased (P<0.01);the expressions of CHOP mRNA and protein increased (P<0.01). Compared with the model group, FBG and UmAlb in each treatment group significantly decreased (P<0.01);the expressions of GRP78 mRNA and protein in the ZGJTYS group and 4-Phenylbutyric acid group increased (P<0.01), the expressions of CHOP mRNA and protein decreased (P<0.01). The renal pathological lesions of each treatment group were significantly improved compared with model group. Conclusion ZGJTYS Decoction can effectively improve the damaged podocyte induced by endoplasmic reticulum stress, delay DN progress.

ABSTRACT

Objective To investigate the effects of Zuogui Jiangtang Yishen decoction (ZGJTYS) on the expression of nephrin and podocin in podocyte of MKR mice with diabetic nephropathy (DN), and explore its mechanism. Methods Forty MKR mice (8 weeks old) were randomly divided into four groups as follows:negative control group (group A), DN model group (group B), ZGJTYS group (group C) and positive control group (group D, Gliquidone and benazepri). All mice from group B, C and D were received high-fat diet feed and unilateral nephrectomy. Four weeks after operation, all mice were received drug intervention, and four weeks later, all mice were put to death. The levels of UmAlb were observed by ELISA, the serum BUN and Cr by biochemical, and the FBG by electrochemical detection method. The nephrin and podocin mRNA expression were measured by RT-PCR and the protein expression by western blotting. The morphological structure changes of the podocytes were observed by transmission electron microscopes. Results As compared with group A, FBG, BUN, SCr and urine UmAlb in the mice of group B increased significantly (P<0.01), the expression level of nephrin and podocin mRNA and protein were markedly decreased (P<0.01). After intervention of drugs, all biochemical indicators above in the mice of group C and D significantly decreased (P<0.01), the expression level of nephrin and podocin mRNA and protein were markedly increased (P<0.01, P<0.05), compared with group B. The renal pathological lesions of group C and D were significantly improved compared with group B. Conclusion ZGJTYS decoction exerts reno-protective effect via reinstating nephrin and podocin expression to repair the damaged podocyte.

ABSTRACT

Objective To investigate the protective effects of Jiangtangyishen Decoction on early-stage diabetic nephropathy of MKR mice. Methods Twenty-four MKR mice were randomly divided into three groups after stratification by sex and fasting plasma glucose, i.e. the model group, traditional Chinese medicine (TCM) group with Jiangtangyishen Decoction 16.72 g/(kg?d), western medicine group with Gliquidone 4.33 mg/(kg?d) and Benazepril 1.44 mg/(kg?d). In the same period, the C57BL/6 wilding mice were the normal group. After treated for 30 days, the levels of blood glucose, the content of microalbuminuria and ?2-microglobulin in urine were observed by ELISA. Renal pathologic lesion of each group were observed by light microscopy and electron microscopy.
